This work is to investigate expression changes of APE/Ref-1 in human pancreatic cancer cells irradiated with colloid of chromic 32P phosphate (32P-CP). Human pancreatic cancer cell line of Patu-8988 was irradiated with 0 0.11

0.45 or 0.90 MBq/mL of 32P-CP for 24 h. The mRNA and protein levels of APE/Ref-1 were determined by RT-PCR and Western-blot respective. The results show that the expression of APE/Ref-1
both of mRNA and protein levels were elevated significantly and were positively related to the irradiation dose. These suggested that elevated APE/Ref-1 expression might be an adaptive response contributing to the overall radioresistance seen in pancreatic carcinomas and implied the potential theraputic effects of gene silence targeted to APE/Ref-1.
